Activation of oligodendroglial Fyn kinase enhances translation of mRNAs transported in hnRNP A2–dependent RNA granules
Central nervous system myelination requires the synthesis of large amounts of myelin basic protein (MBP) at the axon–glia contact site. MBP messenger RNA (mRNA) is transported in RNA granules to oligodendroglial processes in a translationally silenced state. This process is regulated by the trans-acting factor heterogeneous nuclear ribonucleoprotein (hnRNP) A2 binding to the cis-acting A2 response element (A2RE). Release of this repression of MBP mRNA translation is thus essential for myelination. Mice deficient in the Src family tyrosine kinase Fyn are hypomyelinated and contain reduced levels of MBP. Here, we identify hnRNP A2 as a target of activated Fyn in oligodendrocytes. We show that active Fyn phosphorylates hnRNP A2 and stimulates translation of an MBP A2RE–containing reporter construct. Neuronal adhesion molecule L1 binding to oligodendrocytes results in Fyn activation, which leads to an increase in hnRNP A2 phosphorylation. These results suggest that Fyn kinase activation results in the localized translation of MBP mRNA at sites of axon–glia contact and myelin deposition

Toxic mercury is still being used today for example by workers mining gold, resulting in diverse health symptoms in users and individuals in proximity. A considerable burden of disease (BoD) can be assumed, while previous analyses were limited by data scarcity. Besides limited epidemiological data, neither data about the health-related quality of life (HRQoL) nor about the disease severity (disability weight, DW) is available. The aim of the project was to develop disease profiles of chronic metallic mercury vapor intoxication (CMMVI) by including the HRQoL to improve the data basis for BoD analyses of gold miners exposed to mercury.
Methods
Disease profiles comprising the disease label [a], differentiation into disease stages [b], description of the cause of exposure [c], a list of common symptoms [d], and an assessment of the HRQoL [e] were developed using expert elicitation and literature search. The HRQoL was assessed by experts using the five EuroQol dimensions accompanied by the cognition add-on questionnaire (EQ-5D + C).
Results
The ten sources used for the analyses (interview transcript, presentation, and eight literature reviews) identified more than 250 terms describing 85 distinguishable health effects of CMMVI. The analysis revealed 29 common symptoms that were frequently mentioned. Moderate and severe CMMVI cases differ regarding their symptoms and/or symptom severity and HRQoL, resulting in the EQ-5D + C-3L codes 121222 and 233333, respectively.
Conclusions
The profiles should be used to facilitate the ascertainment of CMMVI cases, to compare the HRQoL with other diseases, to derive DWs for improving BoD estimates, and to foster discussions about how to reduce the associated burden

Qualitätsentwicklung an Ganztagsschulen
Durch die Verlagerung bzw. Stärkung von Entscheidungskompetenzen auf die bzw. der Ebene der Einzelschule wird es ermöglicht, Lösungs-/Gestaltungsansätze zu entwickeln, die auf die jeweiligen Bedürfnisse und Gegebenheiten vor Ort zugeschnitten werden können. Die kritische Auseinandersetzung mit den Erfahrungen anderer, die auf entsprechenden Fortbildungsveranstaltungen kommuniziert werden können, lässt Good-practice-Beispiele entstehen, aus denen sich Anregungen zur Realisierung eigener Vorhaben im Zuge der Ganztagsschulentwicklung ableiten lassen.
Der dritte bayerische Ganztagsschulkongress "Qualitätsentwicklung an Ganztagsschulen" am 1. und 2. März 2012 in Forchheim bot den Teilnehmerinnen und Teilnehmern anhand diverser Vorträge, Workshops und Schulbesuchen die Möglichkeit zu Diskussion und Austausch. Developmental molecular factors of the dentate gyrus in mice
Der Gyrus dentatus bietet ein gutes Modell zur Untersuchung diverser, die
Kortexentwicklung betreffender Fragestellungen, insbesondere solche bezüglich
Neurogenese, neuronaler Migration, Arealisierung, axonaler Wegfindung und
neuronaler Plastizität. Weiterhin besitzt er die außergewöhnliche Fähigkeit,
lebenslang funktionelle Neurone aus neuronalen Vorläuferzellen zu bilden, was
ihn zu einem attraktiven Forschungsobjekt für mögliche neuronale
Regenerationsprozesse im Kortex macht. Das Ziel dieser Arbeit war, molekulare
Faktoren der Gyrus dentatus Entwicklung zu identifizieren und ihre Funktionen
zu analysieren. Gegenstand der Experimente war ein Vergleich der
Expressionsmuster von Emx2-/- Mutante gegen Wildtyp Maus, da der
Transkriptionsfaktor Emx2 als Schlüssel-Regulator der hippocampalen
Entwicklung fungiert und sein Fehlen eine sehr spezifische Veränderung dieser
verursacht, was unter anderem zur Folge hat, dass ein morphologisch
erkennbarer Gyrus dentatus in dieser Mutante nicht vorhanden ist. Es wurden
bekannte Gene, deren Funktionen in der Literatur bereits umfangreich
beschrieben worden sind, mit Hilfe von In Situ Hybridisierungen im
Entwicklungsverlauf untersucht. Von besonderem Interesse war hierbei die
Analyse der verschiedenen Funktionskreise wie Proliferation, Differenzierung
und Migration sowie der strukturellen Veränderungen des Gyrus dentatus und der
an diesen angrenzenden Strukturen (hippocampale Fissur und Marginalzonen).
Zudem wurden Kandidatengene, die bisher nicht mit Emx2 in Zusammenhang
gebracht worden sind, durch Screening von Mikroarrays ermittelt. Die
Ergebnisse zeigen, dass die proneurale Kaskade der bHLH Gene im Emx2-/-
Mutanten Gyrus dentatus zwar vorhanden, aber abnorm auf die germinative Zone
beschränkt ist, migrierende Vorläufer der Neurone und der Glia fast völlig
fehlen, das Gerüst der radialen Glia defekt ist und die geringe Anzahl der
Astrozyten und Körnerzellen, die produziert werden, nicht fähig sind, bis zu
ihrem eigentlichen Ziel zu wandern. Zudem erfolgt keine vollständige
Ausdifferenzierung der Körnerzellen. Auch im Bereich der sich entwickelnden
Fissur fehlt bei der Emx2-/- Mutante das Gerüst der radialen Glia, die innere
Marginalzone (IMZ) scheint normal entwickelt zu sein, die äußere Marginalzone
(OMZ) und die Marginalzone des Gyrus dentatus (DMZ) hingegen sind atrophisch.
Die Reln-exprimierenden Zellen dieser Zonen fehlen, mit Ausnahme einer
spezifischen Subpopulation, die mit Hilfe zusätzlicher Marker charakterisiert
wurde, vermutlich aus dem „Hem“ stammt und Emx2 offensichtlich nicht benötigt.
Die Mikroarray Analyse lieferte eine Gruppe von Genen, deren Expression im
Emx2-/- Hippocampus reduziert ist und die zum großen Teil bisher noch nicht
mit Emx2 in Verbindung gebracht wurden. Einige der Kandidaten sind Gene der
Gehirnentwicklung oder solche, die an der funktionellen Reifung beteiligt sind
und werden in der hippocampalen Platte und/oder dem sich entwickelnden Gyrus
dentatus exprimiert. Eine zweite Gruppe von Kandidaten markiert neuronale,
gliale oder vaskuläre Strukturen in der OMZ und repräsentiert wahrscheinlich
Marker für Zellpopulationen, die in der Emx2-/- Mutante spezifisch fehlen. Die
Abwesenheit dieser Gene in der Emx2-/- Mutante weist auf Veränderungen in
verschiedenen Pfaden und Prozessen hin, insbesondere im Notch- und Chemokin-
Pfad, in Prozessen der Zellmigration, der axonalen Wegfindung und der
Angiogenese; zudem wird ein Zusammenhang zwischen der hippocampalen
Morphogenese und der Pia sowie der Cajal-Retzius Zellen aufgezeigt.The dentate gyrus provides a good model for diverse areas of research
concerning the cortical development, especially neurogenesis, neuronal
migration, regionalization, axonal guidance and neuronal plasticity.
Furthermore, the dentate gyrus has the special ability to produce functional
neurons out of neuronal precursors the whole life long. Therefore it is an
interesting object of research for potential neuronal processes of
regeneration in the cortex. The aim of this study was to identify molecular
factors of dentate gyrus development and to analyse their functions. The
subject of the experiments was a comparison of expression patterns from
Emx2-/- mutant versus wildtype mouse, because the transcription factor Emx2
plays a key role in hippocampal development. Thus, mice deficient in this
factor show very specific developmental alterations in the hippocampus, which
leads to the absence of a morphologically visible dentate gyrus in these
Emx2-/- mutants. First some known genes, whose functions had already been
described in literature extensively, were analysed during development by in
situ hybridisation. Thereby investigating the different functional circuits
like proliferation, differentiation and migration as well as the structural
alterations of the dentate gyrus and the adjacent hippocampal fissure and
marginal zone were of particular interest. Furthermore some candidate genes,
for which a putative relation to Emx2 had not been described previously, were
identified by screening microarrays. The results show that, in the Emx2-/-
mutant dentate gyrus, the proneural cascade of bHLH genes is present, but
abnormally confined to the germinal layer, the neuronal and glial migrating
precursors are nearly absent, the glial scaffold is defective, the few
produced astrocytes and granule cells are not able to migrate to their proper
destination and in addition the granule cells are deficiently differentiated.
The glial scaffolding is also lacking in the Emx2-/- mutant developing
hippocampal fissure. The inner marginal zone (IMZ) seems to be developed
properly, in contrast the outer marginal zone (OMZ) as well as the dentate
marginal zone (DMZ) is atrophic. Most of the Reln-expressing cells that should
occupy the OMZ and DMZ are missing, except a specific subpopulation,
characterized by using additional markers, probably born in the hem and for
which Emx2 is not essentially required. The microarray analysis reveals a
group of genes whose expression is reduced in the Emx2-/- hippocampus. For
most of them a putative relation to Emx2 pathways had not been recognized
previously. Some candidates are development genes or are involved in
functional maturation and show expression in the hippocampal plate and/or
developing dentate gyrus. A second group of candidates labels neuronal, glial
or vascular structures in the OMZ and likely represent markers for cell
populations specifically absent in the mutant. The lack of these genes points
at different pathways and processes altered in the Emx2-/-mutant, particularly
the Notch and chemokine pathways, the processes of cell migration, axonal
guidance and angiogenesis, also a relation of hippocampal morphogenesis with
pia and Cajal-Retzius cells is shown
